Assessing the integrity and permeability of barriers is crucial for understanding barrier-forming tissues. Real-time monitoring of barrier permeability and integrity leverages impedance spectroscopy, a widely employed and potent technique. Comparatively, analyzing data collected from different devices is deceptive because of the emergence of a non-homogeneous field across the tissue barrier, substantially complicating impedance data normalization. To monitor barrier function, this work incorporates PEDOTPSS electrodes and impedance spectroscopy, resolving this issue. Across the entire expanse of the cell culture membrane, a homogenous electric field is created by semitransparent PEDOTPSS electrodes. Consequently, each section of the cell culture area is equitably represented in the measured impedance. According to our present knowledge, PEDOTPSS has never been used independently to monitor the impedance of cellular barriers while simultaneously enabling optical inspections within out-of-cell conditions.
